Abstract
The oligosaccharide tag I from our lab combines UV-activity for detection and bioaffinity for purification.1 However, the labeling efficiency of I was low for higher molecular weight oligosaccharides, presumably due to steric hindrance by the biotin moiety. The labeling efficiencies of compounds II and III, respectively, suffered from reduction of the nitro group (II) and steric hindrance by the amide, methoxy and biotin group (III). With amines IV-VI, we observed increased labeling efficiency with increasing length of the spacer. Based on these results, two new tags VII and VIII were synthesized. These tags proved highly efficient in case of smaller sugars. Labeling of higher molecular weight sugars was investigated as well.
